Mendoza impersonated Kris Aquino in a Dubsmash collection she shared on Facebook in the early months of 2015, which quickly gained one million views. She was given the nickname "Queen of Dubsmash" in the Philippines as a result. She was subsequently chosen to play Yaya Dub, better known as Divina Ursula Bukbukova Smash, Lola Nidora's personal assistant on Eat Bulaga! first appeared on July 4, 2015, as part of the "Juan For All, All For Juan" segment. In the episode, her character was teamed with an Eat Bulaga! co-host Alden Richards that was performed on July 16, 2015, in the romantic comedy sketch segment of the show dubbed "Kalyeserye," without the use of a script.

A combination of Richards' first name and Mendoza's character from "Kalyeserye," the couple became known as AlDub, which led to their widespread visibility and to a rise in Eat Bulaga's TV ratings. Her first appearance in a real teleserye was when Mendoza joined the cast of Princess in the Palace as Chef Elize in November 2015. She had two cameo appearances in December 2015: one in the Sunday noontime variety show Sunday PinaSaya and one in the TV sitcom Vampire Ang Daddy Ko.

Together with Vic Sotto and Ai-Ai de las Alas, Mendoza and Richards starred in the 2015 Metro Manila Film Festival (MMFF) entry My Bebe Love #KiligPaMore. Mendoza will appear in the movie, according to Jose Javier Reyes, the film's director. Mendoza portrays Vic Sotto's character's obstinate and disobedient daughter, Anna Carillo. The MMFF honored her for her performance by giving her the Best Supporting Actress trophy.

They starred in a Christmas station ID ad for GMA Network in December 2015. Despite the fact that she appears on GMA, she is not a member of the GMA Artist Center, and Triple A, the talent agency run by Eat Bulaga! executive producer Antonio P. Tuviera, is in charge of managing her career.

Mendoza took part in Eat Bulaga's drama specials during the Lenten season. March 2016 She returned to her role as Divina/Yaya Dub in Kaputol ng Buhay and played Dani in God Gave Me You, for which she was nominated for an Eat Bulaga award for Most Dramatic Performance by a Lead Star and received the Darling of the Screen award. 2016's 3rd EB Dabarkads Awards was place on April 2. Imagine You & Me had Mendoza and Richards as its primary actors, and some of the movie's scenes were filmed in Como, Italy.

In the 2017 television series Destined to be Yours, Mendoza and Richards played the love interests of Benjie Rosales, an architect played by Richards. Mendoza played the part of Sinag Obispo, a radio DJ, in the show. Mendoza and Richards were the stars of an Eat Bulaga! Love is a made television movie... with director Adolf Alix Jr. Since December 2017, Mendoza has frequently made appearances in Daig Kayo ng Lola Ko in various guest roles.
